Malaria
Malaria is a parasitic disease that involves high fevers, shaking chills, flu-like symptoms, and anemia.
Causes
Malaria is caused by a parasite that is transmitted from one human to another by the bite of infected Anopheles mosquitoes. In humans, the parasites (called sporozoites) travel to the liver, where they mature and release another form, the merozoites. These enter the bloodstream and infect the red blood cells. The parasites multiply inside the red blood cells, which then rupture within 48 to 72 hours, infecting more red blood cells. The first symptoms usually occur 10 days to 4 weeks after infection, though they can appear as early as 8 days or as long as a year after infection. Then the symptoms occur in cycles of 48 to 72 hours. The majority of symptoms are caused by the massive release of merozoites into the bloodstream, the anemia resulting from the destruction of the red blood cells, and the problems caused by large amounts of free

This web resource provides data and information relevant to malaria genetics and genomics. These resources include organism specific sequence BLAST databases (Plasmodium falciparum only, all Plasmodium ), genome maps, linkage markers, and information about genetic studies. Links are provided for other malaria web sites and genetic data on related apicomplexan parasites. MIMCom, a project of the Multilateral Initiative on Malaria and the National Library of Medicine , was conceived by African malaria researchers in 1997 and designed and implemented by NLM in collaboration with partners in Africa, US, UK, and Europe. The mandate for Internet access to medical literature came from African scientists: "Access to e-mail and the Internet will promote rapid communication between investigators working at different sites as well as access to online literature and data available to scientists outside Africa." Contact The malaria vaccine programme at Oxford is a linked pre-clinical and clinical vaccine development programme that has primarily targeted the induction of high level T cell responses against pre-erythrocytic antigens. We undertook the first prophylactic DNA vaccine trials in Europe and showed that DNA administered by needle or by gene gun generated only moderate immunogenicity and was not protective. Following on from extensive pre-clinical studies we undertook the first clinical trials of any recombinant MVA vaccine and found a recombinant expressing the ME-TRAP insert more immunogenic than DNA, but this was not protective used alone. However, prime-boost regimes with DNA followed by MVA were substantially more immunogenic with T cell responses reaching 1000 SFU/million.
A fowlpox (FP9)-MVA prime-boost regime was then tested in phase I/IIa clinical trials and induced good T cell immunogenicity with a higher proportion of CD8 T cells. Significant delays in time to parasitaemia were observed with both DNA-MVA and FP9-MVA regimes with an estimated 80-90% reduction in parasite load at the liver-stage. Stronger T cell responses correlated with better protection levels, but only occasional individuals showed sterile protection pointing to a need for more potent vaccines.
