Cryobiology: The Study of Life and Death at Low Temperatures
by Gregory M. Fahy, PhD Cryobiology usually thought of as the study of the effects of subfreezing temperatures on biological systems stands at the interface between physics and biology. The physical principles of cryobiology are universal, which provides some coherence to the field, but the biosphere contains many surprises and twists, which adds endless fascination. The present very abbreviated overview is primarily directed at living systems exposed to temperatures below zero degrees Celsius, but it should be printed out that the cryobiology tent covers all branches of low-temperature biology, including the effects of temperatures above freezing. Technically, cryobiology is actually the study of living systems at any temperature below the standard physiological range. This includes, for example, human hypothermia (both deliberate and accidental) and even natural hibernation, which is a physiological modification of sleep that has allowed the physiological temperature range to be stretched to include temperatures that were previously fatal. Above-freezing temperatures can be just as lethal as sub-freezing temperatures, a fact that has significant ecological and agricultural significance.

Department of Applied Physics RMIT Univeristy , Melbourne Low temperature damage in cells can be divided into damage produced by three effects: (i) low temperature per se ; (ii) direct effects of freezing and (iii) indirect effects of freezing . Cryobiology (the study of life at low temperatures) and anhydrobiology (the study of life at low water contents) have some features in common. This is because, in environmental freezing, one of the major causes of damage is freezing induced dehydration. This essay gives an introduction to several types of cryobiological and anhydrobiological damage at the cellular level. It concentrates however on freezing- or desiccation-induced cellular dehydration, and on our biophysical research in this area and on related systems. References to related scientific papers are given at the end.

    Introduction
    The study of damage produced by freezing and/or low temperatures and by low water contents is important in a variety of fields, of which here are some examples: In medicine, surgeons would like to be able to cryopreserve organs for transplants. To date, however, the cryopreservation of large organs (except blood) has a very poor success rate. Blood and sex cells are routinely frozen and thawed for later use but even then, in many cases, the cellular survival rates are unacceptably low. Cryopreservation is also important in maintaining germplasm for important or endangered species. Frost damage is an important agronomic concern: if farmers can get a crop into the ground before the last frost, then they have a longer growing season and a greater yield. Damage in seeds during drying and rehydration may also be agronomically and ecologically important. Drying and freeze drying are also important in the food industry.

Cryobiology
the study of the physiology of low temperature.
Prior to the advent of modern medical science, it had been observed that survival from severe hypothermia was possible, if uncommon. Many animals enter states of dormancy to conserve water and energy during particularly adverse climatic conditions e.g., dormice and bears, lungfish and some desert toads on Terra. However, there is a limit to how long dormancy can be maintained. Metabolism continues, albeit at a greatly slowed rate. Long term tissue preservation requires metabolic arrest. To effect this, cell water must be locked in place to prevent cellular proteins from working. Water unfortunately expands on freezing (crystallisation), which causes cell rupture and death. It had been observed that rapid freezing limited the size of ice crystals formed. Immersion in liquid nitrogen (-196°C) was used to preserve sperm, ova, embryos, seeds and pollen as early as Tech Level 6. By Tech Level 8, it had been shown that animals such as frogs and mice could be snap-frozen, thawed and revived. Survival rates were predictably low. The great breakthroughs which usher in the modern era of low berthing take place at Tech Level 9.

 Background
CRYO -  (Greek: kryos) cold, frigid (Citation 20) Cryobiology  is the branch of biology that studies the effects of low temperatures on living things. Understanding how to manipulate the freezing process is the cryobiologist’s ultimate goal. Just as food is stored in the deep freezer, low temperatures have the ability to preserve life. On the other hand, these temperatures also have the ability to destroy. Frozen crops or frostbite on ears or toes are examples where the cold has damaged cells so badly that they are destroyed. It is this dual nature of the cold to preserve and destroy that has created its own special field of cryobiology in the entire subject of biotechnology and to subfields such as cryopreservation and cryosurgery.  (Citation 8) Above: Because of evolution, some organisms have adapted to survive at extremely low temperatures, even below the freezing point of water (0°C). Their ability to create biomolecules allow these organism to prevent the fluid in their cells from freezing solid. The figure above demonstrates the wide tempreture range that of living organism. The Himalayan midge,an insect found in glaciers, is still active at -20°C. (Citation 8) Above: Cell and tissue banks of frozen cells help scientists manufacture cellular products and conduct experiments on cells. Keeping their samples at cold tempratures allows scientists to keep desirable cells in a state of suspended animation. Scientists most commonly freeze thin tissue cells like skin cells, but the extention of this procedure has been known to include the successful freezing and transplant of organs. (Citation 8)

[Repeat if necessary] What is cryonics? Cryonics Cryonicists are people who use or advocate cryonics in the hope of greatly extending life and youth. Most diseases, including the progressive deterioration known as "getting old", are the result of damage to organs, tissues, cells and cellular components. With enough progress of medicine and molecular repair capability, all diseases should eventually be curable, including aging. Medicine in the future should be able to restore and maintain people in a condition of youth and health. Cryonics could be a lifeboat (or "first aid") to future medicine. Because it is based on speculation about the capabilities of future science cryonics is not a science. Few scientists are qualified to say whether future science can or cannot realize the dream that motivates cryonics because scientists are only trained in current science. Many outstanding scientists have made false predictions about future technology. In 1885 Lord Kelvin declared that "Heavier-than-air flying machines are impossible." A couple of decades ago nearly all scientists believed that cloning is impossible. Conversely, cryonicists cannot guarantee that cryonics will work. Only the future will tell whether the predictions of cryonicists are correct. If cryonics works and a person has not been cryopreserved, that person has no chance of ever being restored to an enduring youthful and healthy condition. If cryonics does not work at any moment in time, it may be made to work in the future. Those who have been cryopreserved will simply wait.
