Yi Pan SecA is an important component of protein translo- cation in bacteria, and exists in soluble and membrane-integrated forms. Most membrane prediction programs predict SecA as being a soluble protein, with the exception of TMpred and Top- Pred. However, the membrane associated predicted segments by TMpred and TopPred are inconsistent across bacterial species in spite of high sequence homology. In this paper we describe a new method for membrane protein prediction, PSSM_SVM, which provides consistent results for integral membrane domains of SecAs across bacterial species. This PSSM encoding scheme demonstrates the highest accuracy in terms of among the common prediction methods, and produces consistent results on blind test data. None of the previously described methods showed this kind of consistency when tested against the same blind test set. This scheme predicts traditional transmembrane segments and most of the soluble proteins accurately. The PSSM scheme applied to the membrane-associated protein SecA shows characteristic features. In the set of 223 known SecA sequences, the PSSM_SVM prediction scheme predicts eight to nine residue embedded mem- brane segments. This predicted region is part of a 12 residue helix from known

Abstract This paper provides evidence that normal animal cells possess a unique regulatory mechanism to shift them between proliferative and quiescent states. Cells cease to increase in number under a diversity of suboptimal nutritional conditions, whereas a uniformity of metabolic changes follows these nutritional shifts. Evidence is given here that cells are put into the same quiescent state by each of these diverse blocks to proliferation and that cells escape at the same point in G of the cell cycle when nutrition is restored. The name restriction point is proposed for the specific time in the cell cycle at which this critical release event occurs. The restriction point control is proposed to permit normal cells to retain viability by a shift to a minimal metabolism upon differentiation in vivo and in vitro when conditions are suboptimal for growth. Malignant cells are proposed to have lost their restriction point control. Hence, under very adverse conditions, as in the presence of antitumor agents, they stop randomly in their division cycle and die.

Abstract The minor form of valine tRNA from baker's yeast-tRNAVal 2bpurified by column chromatography was completely digested with guanylo-RNase and pancreatic RNase. The products of these digestions were separated by a combination of thin-layer chromatography on cellulose and high voltage electrophoresis on DEAE-paper and then identified. The halves of tRNA Val 2b were prepared by partial digestion with pancreatic RNase, and their complete guanylo-RNase and pancreatic RNase digests were analysed.
