Schally, Andrew V.; Bajusz, Sandor; BACKGROUND OF THE INVENTION
The present invention relates to novel peptides which inhibit the release of gonadotropins by the pituitary gland in mammals without inducing edematous reactions. More specifically, the present invention relates to analogs of the luteinizing hormone releasing hormone (LHRH), which has the structure:
p-Glu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH.sub.2,
salts thereof, and to pharmaceutical compositions and methods of use pertaining to these analogs.
DISCUSSION OF THE PRIOR ART
For more than 15 years, investigators have been searching for selective, potent antagonists of the LHRH decapeptide (M. Karten and J. E. Rivier, Endocrine Reviews, 7, 44-66 (1986)). The high degree of interest in such antagonists is due to their usefulness in the fields of endocrinology, gynecology, contraception and cancer. A large number of compounds have been prepared as potential LHRH antagonists. The most interesting antagonists to date have been compounds whose structure is a modification of the structure of LHRH.
The first series of potent antagonists was obtained by introduction of aromatic acid residues into positions 1, 2, 3 and 6, or, 2, 3, and 6. The compounds are expressed as LHRH modified by replacement of the original amino acid residues by others at the position indicated by the superscript numbers. The known antagonists include:

    hypothalamus u u m u s) [ key , an important supervisory center in the brain , rich in ganglia, nerve fibers, and synaptic connections. It is composed of several sections called nuclei, each of which controls a specific function. The hypothalamus regulates body temperature, blood pressure, heartbeat, metabolism of fats and carbohydrates, and sugar levels in the blood. Through direct attachment to the pituitary gland , the hypothalamus also meters secretions controlling water balance and milk production in the female. The role of the hypothalamus in awareness of pleasure and pain has been well established in the laboratory. It is thought to be involved in the expression of emotions, such as fear and rage, and in sexual behaviors. Despite its numerous vital functions, the hypothalamus in humans accounts for only 1/300 of total brain weight, and is about the size of an almond. Structurally, it is joined to the thalamus ; the two work together to monitor the sleep-wake cycle.

    Po tym wstępie latwo mi będzie przejść do prezentacji człowieka, który w 1977 roku otrzymał nagrodę Nobla w dziedzinie nauk medycznych i oficjalnie przyznaje się do koneksji z Polską.
    Po wojnie pozostał w Szkocji i tam także Andrzej Schally ukończył szkołę średnia, skąd wyjechał do Londynu na studia chemiczne. Pracę naukową rozpoczął w National Institute of Medical Research. Chyba nie odważyłabym się o Andrzeju pisać, bo na początek zaplanowałam prezentację Polaków pracujących dla wspólnego światowego dziedzictwa, a w jakis sposób związanych także z Kanadą, gdyby nie fakt, że właśnie w Kanadzie, w Montrealu podjął pracę w 1952 roku w charakterze pracownika naukowego na Uniwersytecie McGill i właśnie w Kanadzie (jak sądzę) wykształciły się jego zainteresowania endokrynologią. Endokrynologia jest poddyscypliną medyczną dość młodą, ale ma już wielu entuzjastów wierzących głęboko, że zglębianie tejże pomoże ujawnić procesy dotąd może omijane, może lekceważone, ale z pewnością traktowane po macoszemu.

Targeted cytotoxic analog of luteinizing hormone-releasing hormone AN-207 inhibits growth of OV-1063 human epithelial ovarian cancers in nude mice
Article Abstract: Targeting chemotherapy to the receptor for luteinizing hormone-releasing hormone may reduce its toxicity when used to treat ovarian cancer. This is done by combining the chemotherapy drug with luteinizing hormone-releasing hormone, a naturally occurring hormone in the body. Some types of ovarian cancer have the receptor for luteinizing hormone-releasing hormone on their surface. The targeted chemotherapy would bind to this receptor and the toxic drug would kill the tumor. In a study of mice, the hormone was linked to a chemotherapy drug related to doxorubicin (Adriamycin). This combination inhibited tumor growth with few side effects.
