The Application of the Freezing-Drying Technique to Retinal Histochemistry
Description:
In this article, Anfinsen, Lowry, and Hastings presented a method that permitted histological examination and quantitative enzymatic study of individual freeze-dried sections of tissue in order to study enzymatic localization in the various layers of the retina. Published shortly before Anfinsen received his Ph.D. from Harvard in 1943, this is a quantitative, histochemical study of the retina in which the three researchers adapted an earlier technique for the identification of chemical components in cells and tissues that was developed by Kaj Linderstrom-Lang and K.R. Morgan.

Il a entrepris l'édition des Advances in Protein Chemistry, et nous a laissé un important ouvrage: The Molecular Basis of Evolution, paru en 1959.
Ainsi, dès 1962, la preuve était faite que la séquence des résidus déterminée par le code génétique détermine à son tour les autres structures des protéines; résultat d'une importance capitale. Dans le processus de renaturation de la ribonucléase, les huit cystéines (S-H) se réoxydent grâce à l'oxygène atmosphérique, et rétablissent ainsi les quatre ponts disulfure; ceux-ci impliquent les mêmes paires de cystéines que la molécule native. Or, bien que le calcul de probabilités montre que les huit cystéines d'une seule chaîne polypeptidique peuvent former jusqu'à 105 ensembles de ponts di sulfure différents, seul l'ensemble qui était présent dans la molécule de ribonucléase native sera formé.
Au cours des années 1970, Anfinsen et son équipe ont travaillé sur une nouvelle nucléase extracellulaire de staphylococcus aureus. Ils ont déterminé la séquence des 149 amino-acides qui la composent, et ont décrit ses propriétés enzymatiques, chimiques et immunologiques, faisant appel à toutes les techniques modernes de la chimie, de la spectroscopie et de la chromatographie. Pour affiner ses travaux, Anfinsen, en collaboration avec Cuatrecasas, a mis au point le principe de la chromatographie d'affinité, qui rend depuis lors d'immenses services pour la purification et l'identification des cibles hormonales des membranes plasmiques dans les cellules eucaryotiques, et pour l'identification des acides aminés dans une chaîne polypeptidique, ainsi que la détermination de leurs sites actifs.

Antibodies reactive with distinct regions of the staphylococcal nuclease molecule were prepared both by immunization with polypeptide fragments of nuclease and by immunization with intact nuclease followed by fractionation of the antiserum on immuno-absorbent columns bearing the corresponding fragments. Comparisons of the interactions of these antibody preparations with nuclease, by quantitative precipitin assays and enzyme inhibition studies, showed marked differences attributable to the conformation of the immunizing antigens. An interpretive model is proposed in which antibodies fractionated from anti-nuclease serum react effectively only with the ``native format determinants'' of polypeptide fragments of nuclease. It is postulated that such determinants are generated in polypeptide fragments by spontaneous and reversible folding of the polypeptide chain. The model permits experimental determination of parameters, K conf , for the proposed conformational equilibria.

    Christian Anfinsen was born in Monessen, Pennsylvania into a family of Norwegian American immigrants. The family moved to Philadelphia in the 1920s. He earned a bachelor's degree from Swarthmore College in 1937. While attending Swarthmore College he played varsity football and joined the Delta Upsilon Fraternity.
