ACMAL eTraining Introduction
This document is a publication of the Applied Chemical and Morphological Analysis Laboratory (ACMAL) at Michigan Technological University. It is intended to provide refresher and supplementary training for researchers using the ACMAL facilities. ACMAL is a university facility that houses an extensive array of electron microanalytical and X-ray instruments. The Center is managed by the Department of Materials Science and Engineering and is located on the 6th floor of the Minerals and Materials Engineering Building on the Michigan Tech campus.
For whom is this eTraining Module intended?
  • Researchers on campus who have taken or are taking the appropriate courses for learning the instruments. High school and undergraduate students who are learning about materials science and engineering.

The TEAM Project The project was presented to the Office of Basic Energy Sciences in October 2002. As a DOE Project, TEAM is subject to specific rules that control its progress via a series of critical decisions CD0-CD4. CD0 (Mission Need) was granted in May 2004, CD1 (Alternative Selection and Cost Range) was signed in September 2005 after a critical design review in April 2005. The Conceptual Design review was signed in April 2006, and the CD2/3 review (Approval of Project Baseline/Readiness to Execute) was signed in December 2006. Tecnai F30 The FEI Tecnai F30 is a scanning transmission electron microscope (STEM) that forms the centerpiece of the Electron Microscopy Core Facility. The microscope can be operated at the maximum accelerating voltage of 300 kV and is capable of atomic resolution imaging in both TEM and STEM modes. The microscope uses a Schottky field emission gun as its electron source, providing the highest current density and the smallest electron probe-size, which are critical for high spatial-resolution analysis using the scanning mode.The microscope is currently equipped with a high performance CCD camera with 4k x 4k resolution for image recording, an X-ray energy dispersive spectrometer (EDX) for compositional analysis, and STEM detectors for bright-field, annular dark-field, and high-angle annual dark-field (Z-contrast) imaging.

El Microscopio Electrónico de Barrido (MEB) hace parte de los equipos interfacultades de la Universidad de los Andes, y se encuentra ubicado en el Laboratorio de Caracterización MEB.
Dispone de un microscopio JEOL, modelo JSM 6490-LV de última generación que opera en los modos de alto y bajo vacío, tiene integrados cuatro detectores que permiten la obtención de imágenes de electrones secundarios (tridimensionales), imágenes de electrones retrodispersados (topográficas, composicionales y de sombra) e imágenes de mapas de puntos EDS (mapping). El detector de espectroscopía por dispersión de energía (EDS) permite además la realización de microanálisis cualitativos y cuantitativos.
El Laboratorio cuenta también con un sistema de recubrimiento de muestras (oro/carbono), así como una secadora de punto crítico, equipos necesarios para la adecuada preparación de las muestras.

Introduction
Electron microscopy is an imaging technique that uses an electron beam to probe a material. Since the wavelength of an electron is much smaller than the wavelength of visible light diffraction light microscopy . When electrons penetrate a sample, they are diffracted to form a diffraction pattern. This diffraction pattern can be transformed with a lens to obtain the sample image. Electron microscopy finds its widest use in materials science and microbiology.
Instrumentation
The use of electron beams requires that the sample be placed in a vacuum chamber for analysis. An electron beam is produced by applying a high voltage to a hot tungsten filament, and accelerating the emitted electrons through a high electric field, typically 10-100 keV. The electron beam is then focused with magnetic field lenses to a typical spot diameter of 1-100 nm on the sample.

High resolution electron microscopy (HREM)
High resolution electron microscopy (HREM) is a variation of TEM that uses phase-contrast microscopy to achieve a resolution of approximately 0.2 nm, which is sufficient to provide atomic-scale resolution. The phase-contrast method uses the phase difference of multiple diffracted beams in addition to the intensity of the diffracted beams. The lattice image that is reconstructed is valid only when the electron beam has a spatial resolution of approximately 0.2 nm and the sample is thin enough that electrons are not scattered multiple times (less than approximately 10 nm). Under these conditions the reconstructed lattice image represents the true structure.
