What is mitochondrial DNA?
Mitochondria are structures within cells that convert the energy from food into a form that cells can use. Although most DNA is packaged in chromosomes within the nucleus, mitochondria also have a small amount of their own DNA. This genetic material is known as mitochondrial DNA or mtDNA. In humans, mitochondrial DNA spans about 16,500 DNA building blocks (base pairs), representing a small fraction of the total DNA in cells. Mitochondrial DNA contains 37 genes, all of which are essential for normal mitochondrial function. Thirteen of these genes provide instructions for making enzymes involved in oxidative phosphorylation. Oxidative phosphorylation is a process that uses oxygen and simple sugars to create adenosine triphosphate (ATP), the cell's main energy source. The remaining genes provide instructions for making molecules called transfer RNA (tRNA) and ribosomal RNA (rRNA), which are chemical cousins of DNA. These types of RNA help assemble protein building blocks (amino acids) into functioning proteins.

1. Introduction
The mitochondrial genome is indispensable to the cellular and organismal biology of Caenorhabditis elegans . An elaborate cellular machinery is employed to maintain mitochondrial DNA (mtDNA), to express it, and to ensure its inheritance. Because it is maternally transmitted and because of its wide-ranging copy number (it can be present at fewer than 100 to tens of thousands of copies per cell), the genetics of mtDNA are characterized by a number of peculiar and as yet incompletely understood features. The fundamental importance of mtDNA to cellular energy metabolism explains the profound effects mtDNA mutations can have; mitochondria are the major source of reactive oxygen species, which are implicated in premature aging and senescence.

Using a set of seven oligonucleotide probes, restriction fragments containing the mitochondrial cytochrome b (cyt b) gene from C. reinhardtii were isolated from a mitochondrial DNA library. This gene is located adjacent to the gene for subunit 4 of the mitochondrial NADH-dehydrogenase (ND4), near one end of the 15.8-kb linear mitochondrial genome of C. reinhardtii. Research Highlights
Our research centers on the mechanisms that enable the cell to take advantage of the high energetic yield of oxidative phosphorylation (OXPHOS) in spite of the concomitant production of reactive oxygen species (ROS). Defects in these mechanisms lead to energy depletion and oxidative damage, which are increasingly implicated in degenerative disease and aging. Mitochondrial iron homeostasis is vital for OXPHOS maintenance and anti-oxidant protection. Both the heme and iron-sulfur cluster biosynthetic pathways depend on a constant supply of iron to the mitochondrial matrix, where micromolar concentrations of iron must be kept in reduced, available, and non-toxic form. This is a formidable challenge for the cell because the superoxide anion and hydrogen peroxide produced by the respiratory chain favor the iron-catalyzed production of the highly toxic hydroxyl radical. Friedreich ataxia (FRDA) provides a dramatic example of what happens when mitochondria lose their ability to process iron properly. FRDA (

Mitochondrial DNA sequences have value as markers of genetic phylogeny and relationships. For example, studies of mitochondrial genetics definitively reveal a common Mongoloid ancestor for Polynesians and American Indians, including the Haida and the Tongans, the Apache and the Hawaiians, the Inca and the Samoans. Given its unique matrilineal inheritance, mtDNA provides an unobscured view of the early peopling of the World, and by following unique sequence polymorphisms in the mtDNA, we can assess prehistoric migrations without interference by the paternal overlay of genetic exchange with later arrivals. The value of mtDNA in assessing ancestry is now well known. A useful website at Emory University seems to be down at this time, but a sense of the information mtDNA sequencing has revealed is shown in the following schematic, archived here: Paternal mitochondria, seen here lined up like grapes along the flagella of human sperm, are discarded during fertilization.
