Il y a eu, avant Merrifield, nous l'avons vu, d'autres lauréats du prix Nobel à avoir travaillé sur la synthèse des peptides, le premier d'entre eux ayant été Emil Fischer. Mais la date marquante reste l'année 1953 où a été réussie, par Vincent du Vigneaud la synthèse de l'oxytocine. La synthèse d'un peptide nécessite plusieurs étapes successives durant lesquelles s'enchaînent les différents acides animés. Opérer en milieu liquide, comme l'avait fait du Vigneaud, soulève d'énormes difficultés, car il faut à chaque étape isoler et purifier les intermédiaires de la réaction. Merrifield chercha donc à simplifier la méthode de synthèse; c'est en 1963 qu'il publie ses expériences, dévoilant la nouvelle méthode de synthèse des peptides sur support solide. Cette méthode va rapidement dépasser le cadre de la synthèse peptidique, et avoir une portée générale en chimie organique, tout particulièrement dans la synthèse des polymères. Le grand mérite de Merrifield et de ses collaborateurs a été de réaliser un appareil automatique, programmé, qui peut avec des rendements élevés produire de façon répétitive les différentes étapes chimiques permettant l'addition de chaque aminoacide à une chaîne polypeptidique qui croît progressivement. Une séquence programmée de réactions s'effectue dans un seul réacteur, à l'aide de pompes calibrées, avec des réactifs ajoutés automatiquement à partir d'un réservoir: l'appareil tient compte des temps de réaction de chaque étape et, grâce à la "technique en phase solide", sépare les produits de la réaction des substances parasites.
