    Extractions: Host defense is present in many forms. Overall, the Immune Response (IR) can be divided into two major classifications; humoral and cell-mediated. While these responses are not mutually exclusive, they provide distinctly different avenues for dealing with pathogenic organisms or altered host cells. These different responses will be discussed in more detail later. Immune Response Humoral Immunity Cell-mediated Immunity (Antibody) (Cytotoxicity) Some of these responses are specific, others are non-specific. This page will introduce host defense mechanisms by defining some commonly used terms and describing the specific cells and tissues involved in these immune responses. DEFINITIONS: Antigen (Ag): A molecule which elicits a specific immune response when introduced into an animal. More specifically, antigenic (immunogenic) substances are: Structurally complex (proteins are usually very antigenic), Accessible (the immune system must be able to contact the molecule), and Foreign (not recognizable as "self"). Antibody (Ab): A glycoprotein produced in response to an antigen that is specific for the antigen and binds to it via non-covalent interactions. Our Immunology team diagnoses and treats both children and adults who have primary immune deficiency disorders (PIDDs). There are more than 140 of these complex conditions. People with PIDDs are born with immune systems that are not working properly or that are missing needed parts. This makes them more open to serious infection and illness. Seattle Children’s is one of the few places in the world where doctors are both carrying out research and providing treatment for primary immune deficiency disorders. Our experts in immunology are known around the world for their research into new ways to diagnose, treat and cure PIDDs. This allows us to use what we learn to provide the most advanced care for people with PIDDs. We work closely with the Fred Hutchinson Cancer Research Center and the Seattle Cancer Care Alliance to coordinate care for our patients who need bone marrow transplants . Silkworms and chickens - Louis Pasteur The idea that there may be some unseen agent that spread through the human population and caused disease was not new. Several hypotheses had been put forward with this core idea. They were not accepted due again in part because of social immaturity. People were still very fatalistic about contracting disease. They had not developed the understanding that the factor that spread disease could be isolated and identified. The medical establishment was still embryonic. There was no research that might provide some basic evidence to support the hypotheses and identify a pathogenic organism. Several scientific advances had to be made before the idea of pathogens could be widely accepted. What was required was the understanding of germs. There was some evidence provided in the shape of Anton van Leeuwenhoeck's development of the microscope. With the microscope he was able to describe organisms not visible to the naked eye, but this still did not trigger the idea that similar organisms could be the cause of infectious disease. The equivalent of today's epidemiologists began to emerge and studied the dynamics of infections and epidemics. They were able to recognize that such diseases as typhoid and cholera spread rapidly in densely populated areas and could sometimes be traced to a physical source. This led to the use of isolation as a defense against infected people spreading the disease. However there was still no recognition that this may indicate a physical entity as the cause of disease.
