Extractions: The MCIC microscopy laboratory is fully equipped to conduct biological research at the subcellular and ultrastuctural level. We house light and confocal microscopes, two transmission electron microscopes and two scanning electron microscopes. We offer a variety of sample preparation techniques and work closely with investigators to optimize the various experimental procedures. The MCIC personnel provides assistance and/or individual training in the use of the light and electron microscopes, and technical support for electron microscopy sample preparation. In collaborative projects, the MCIC staff helps design light/EM microscopy experiments and assists with the interpretation of the data. The MCIC staff also trains investigators who wish to learn microscopy techniques and conduct their own work (sample preparation and embedding for light or electron microscopy, thin and ultra-thin sectioning, etc.).

Extractions: Fax : (724) 864-9851 SCANNING ELECTRON MICROSCOPY (SEM): INSTRUMENTATION: Visualization of material can yield many answers to questions regarding material behavior and process problems. Combined with the instrument's ability to perform particle size analysis, SEM becomes a supplement to other characterization methods. Although more expensive than other methods, the ability to "see" material and determine aspect ratios or other shape factors may prove to be of great advantage to a client. RESULTS: Photomicrographs are provided at magnifications directed by the client. Preparation and loading of the samples are also at the direction of the client (if known). Resulting images may be e-mailed or copied to a CD. TRANSMISSION ELECTRON MICROSCOPY (TEM): Several preparation techniques can be employed when preparing samples for TEM analysis. The resulting sample is very thin, with thickness in the nanometer range, to allow electrons to transmit through the sample. The wavelengths of the electron beams used in TEM to irradiate samples are smaller than the light beams used in light microscopy; therefore, the resolution of TEM is much greater than that of light microscopy. IMPORTANCE: TEM is used to view cell structures, to determine grain sizes, to determine crystallography of materials, and to analyze processes and failures.

Extractions: Introduction The Atomic Force Microscope (AFM) is being used to solve processing and materials problems in a wide range of technologies affecting the electronics, telecommunications, biomedical, chemical, automotive, aerospace, and energy industries. The materials under investigation include thin and thick film coatings, ceramics, composites, glasses, synthetic and biological membranes, metals, polymers, and semiconductors. The AFM is being applied to studies of phenomena such as abrasion, adhesion, cleaning, corrosion, etching, friction, lubrication, plating, and polishing. The real-world examples presented on our web page indicate the breadth of AFM applications for problem solving; yet they represent only a fraction of our experience at Advanced Surface Microscopy. Technical Capabilities AFM images show critical information about surface features with unprecedented clarity. The AFM can examine any rigid surface, either in air or with the specimen immersed in a liquid. "Minor" (and major) differences between "smooth" surfaces are shown dramatically. On one hand, the AFM can resolve very tiny features, even single atoms, that were previously unseen. On the other hand, the AFM can examine a field of view larger than 125 microns (0.005 inch), so that you can make comparisons with other information, e.g. features seen in the light microscope or hazes seen by eye. The AFM can also examine rough surfaces, since its vertical range is more than 5 microns. Our analytical reports of AFM results include three-dimensional images and quantitative data analysis (such as feature sizes, surface roughness and area, and cross-section plots), integrated and interpreted in the context of your problem.

Contacts Brewster Angle Microscopy is a technique that allows the in situ study of thin films at the gas/liquid or solid/gas interfaces. The basis of this method is illustrated at the following scheme (the Brewster angle): For a beam of p-polarised light (p- parallel to the plane of incidence) there is an angle of incidence theta at which no relfection occurs. This is called "Brewster angle" and is satisfying the relationship tan( theta ) = n subphase / n air , where n is the refractive index of the corresponding phase. Introducing a thin film in between the two phases the optical properties of the system change so that a small amount of the incident intensity is reflected. In the general case it can be said that the reflected light can have a different from the incident light polarisation state. The reflected light can easily be "captured" by a detector like a CCD camera and in that way the film can be visualised. top of the page 2. The apparatus
